Trial design
How to plan a useful papain process trial
A practical framework for turning enzyme activity, process conditions and product targets into a measurable comparison.

Begin with a measurable endpoint
Define what success means before choosing the enzyme input. Texture, soluble protein, turbidity, viscosity, yield or a sensory attribute can be useful endpoints when the test method and acceptance range are clear.
Change one system deliberately
Use an untreated or current-process control, then screen a small range of activity inputs. Keep substrate preparation, mixing, pH, temperature and sampling consistent so the comparison remains interpretable.
Measure the final product
The effect immediately after enzyme contact may not represent the packaged or cooked product. Include the downstream thermal, separation or storage steps that determine customer experience.
Plan scale-up before the winning trial
Plant equipment changes mixing, addition accuracy, heat transfer and residence-time distribution. Record the controls that must transfer with the selected condition, not only the nominal dose.
